Britain Human Lysosomal Elastase CATALYTIC AND IMMUNOLOGICAL PROPERTIES
نویسندگان
چکیده
1. The elastase of human spleen was shown to exhibit endopeptidase activity against azocasein and elastin. 2. Activity against several synthetic substrates was detected, and benzyloxycarbonyl-L-alanine 2-naphthyl ester was found to be a good substrate for routine use. 3. The enzyme showed a broad pH optimum in the range of 8.2-9.2 against azo-casein and the synthetic substrate. 4. The effect of inhibitors on the spleen elastase showed it to be a serine proteinase with a specificity similar to that of porcine pancreatic elastase. 5. Specific antisera were raised against the enzyme, and it was shown to be immunologically identical with the lysosomal elastase ofhuman neutrophil leucocytes.

The Comparative Enzymology of Triosephosphate Dehydrogenase.
The properties of crystalline rabbit muscle (1) and yeast (2) triosephosphate dehydrogenases have been studied extensively (3). Velick and Udenfriend (4) have compared some of the properties of the two enzymes. They reported that, although the two enzymes have similar molecular weights and similar amino acid distributions with only small variations, there are differences in the internal structu...

متن کاملProstasin is a novel human serine proteinase from seminal fluid. Purification, tissue distribution, and localization in prostate gland.
A novel serine proteinase, designated as prostasin, has been purified from human seminal fluid to apparent homogeneity by DEAE-Sepharose CL-6B and aprotinin-affinity chromatography. The purified protein migrates as two close bands with an apparent molecular mass of 40 kDa on SDS-polyacrylamide gel electrophoresis under reducing conditions. It can be labeled with [14C]diisopropyl fluorophosphate...
متن کاملMyeloperoxidase mediates neutrophil activation by association with CD11b/CD18 integrins.
Recruitment and activation of polymorphonuclear neutrophils (PMNs) reflects a primary immunological response to invading pathogens and has also emerged as a hallmark of vascular inflammation. One of the principal enzymes released upon PMN activation is myeloperoxidase (MPO), a heme protein that not only generates cytotoxic oxidants but also impacts deleteriously on nitric oxide-dependent signal...
متن کاملDistinct structural and catalytic roles for Zap70 in formation of the immunological synapse in CTL
T cell receptor (TCR) activation leads to a dramatic reorganisation of both membranes and receptors as the immunological synapse forms. Using a genetic model to rapidly inhibit Zap70 catalytic activity we examined synapse formation between cytotoxic T lymphocytes and their targets. In the absence of Zap70 catalytic activity Vav-1 activation occurs and synapse formation is arrested at a stage wi...
